Patients with SLE (185) with moderate to severe disease activity and positive interferon (IFN) gene signature were randomised to receive IFN-K or PBO intramuscular injections (on W0, W1, W4 and W12 and W24). At randomisation (D0) and W12, W24 and W36, the IFN gene signature, including the 21 IFN-regulated genes, was evaluated by Affymetrix.

| Extraction protocol |
Total RNA was extracted from PAXgene blood RNA tubes using the QIASymphony SP extraction robot from QIAGEN. The QIASymphony PAXgene blood RNA Extraction kit and PAXgene_RNA protocol were used.

100 ng total RNA was labeled using the NuGen WB Solution kit according to the manufacturer's instructions.

| Hybridization protocol |
Following fragmentation and biotin labeling, 4.4 µg of cDNA were hybridized for 16 hours at 45C on GeneChip Human Genome U133 Plus 2.0 array. GeneChips were washed and stained in the Affymetrix GeneChip Fluidics Station 450.

GeneChips were scanned using the Affymetrix Gene Chip Scanner 3000.
