TSLP-stimulated CD4+ T cells block the promotion of breast cancer in a spontaneous PyMT mouse model. The goal of this project is to investigate what are the epigenetic changes induced by CD4+ T cells that lead to diminished proliferation of cancer cells Overall design: K14-TSLPtg;PyMT;Rag1KO, and PyMT;Rag1KO mice were injected with wt CD4+ T cells at 4 weeks of age. Tumors were collected when they reached terminal size and tissue was processed for ChIP seq analysis.

Construction protocol: Chromatin extracted from formalin-fixed cells was fragmented with a Branson 250 sonifier, solubilized and immunoprecipitated with H3K27ac antibody (Active Motif, Inc., Carlsbad, CA) overnight at 4°C. Antibody-chromatin complexes were pulled down with protein G-Dynabeads (Life Technologies), washed, and eluted. Immunoprecipitated DNA was extracted with AMP Pure beads (Beckman Coulter) after crosslink reversal, RNase A, and proteinase K treatment. ChIP DNA was quantified with Qubit.
