We have previously reported that the cardenolide drug digitoxin blocks TNFa/NF?B-driven proinflammatory signaling. We therefore hypothesized that digitoxin might break the collaborative process between NF?B and TGFß by also inhibiting expression of TGFBR2. We therefore tested whether TGFß-driven EMT and resulting metastases would be suppressed. Overall design: Digitoxin-treated and vehicle-only treated rats were stratified on the basis of serum IL-6 and serum CINC-1. High serum marker control animals and low serum marker digitoxin-treated animals were used as subjects for differential expression profiling of micropunched primary tumor tissue by mRNA-seq profiling and analysis.

Construction protocol: Samples of grossly dissected, frozen primary tumors were mounted on a cryostat chuck. Serial thick sections (300 micron) were then cut and mounted on cold slides. Ten random samples were micro-dissected from viable regions of the tumor in 300 micron thick sections with a 1 mm diameter micropunch device. The tumor samples were then soaked in RNA Later at four degrees Celsius overnight before isolation of total RNA. One microgram of total RNA was input for library preparation using the TruSeq RNA Sample Prep Kit v2
