Study: The RNA-Binding Protein DND1 Acts Sequentially as a Negative Regulator of Pluripotency and a Positive Regulator of Epigenetic Modifiers Required for Germ Cell Reprogramming

Analysis of our transcriptome and RNA immunoprecipitation experiments indicate DND1 acts as a positive regulator of chromatin modifiers in male germ cells, linking RNA binding proteins and epigenetic regulation. Overall design: Transcriptome timecourse (E12.5, E13.5 and E14.5) comparing muring male germ cells homozygous for Ter mutation in Dnd1 to wild type male germ cells done in conjunction with in vitro work with HEK293 cells transfected with tagged Dnd1 for digestion optimized RNA immunoprecipitation sequencing (DO-RIP-seq) experiments to identify a RNA recognition element for DND1 as well as transcipt targets of this RNA binding protein.
