Study: Dynamic gene regulatory networks of human myeloid differentiation [RNA-seq_siRNA]
show Abstracthide Abstract
We utilize gene expression and open chromatin footprinting data to build a gene regulatory network of key transcription factors that capture the cell and time-specific regulatory programs specified during human myeloid differentiation. Overall design: RNA-seq profiling of undifferentiated HL-60, differentiating macrophage, neutrophil, monocyte, following siRNA control and siPU.1 treatment for 24 hours.
Sample: Mon-24h-siPU Rep1 RNA-seq

Organism: Homo sapiens
Library:
Instrument: NextSeq 500
Strategy: RNA-Seq
Source: TRANSCRIPTOMIC
Selection: cDNA
Layout: PAIRED
Construction protocol: RNA quality was evaluated for all samples using an Agilent 2100 Bioanalyzer. Samples with a RIN > 9 were used to build RNA-seq libraries from 50,000 cells using the Smart-seq2 protocol (Picelli et al., 2013). Reads were sequenced as 40 bp paired-end dual indexed reads using Illumina’s NextSeq 500
