RNA sequencing was used to characterise PGE2-mediated changes in the gene expression profile of in vitro differentiated and repetitively activated antigen-experienced TCF1+CD8+ T cells. Overall design: In vitro differentiated and repetitively activated antigen-experienced TCF1+CD8+ T cells were pre-incubated with or without PGE2 (100 ng/mL) for 1 hour at 37°C. Subsequently, the cells were stimulated for an additional 4 hours with low dose IL-2 (85 U/ml) or the combination of low dose IL-2 and aCD3/CD28 beads.
