To investigate the liver transcriptomics of WT and liver Cullin 3 KO mice on chow diect (CD) and Westrn diet (WD) Overall design: C57BL/6 Cul3 flox mice were on CD or WD for 13 weeks, then AAV-TBG-Null or AAV-TBG-Cre were injected to these mice to generate WT and liver Cul3 KO mice, respectively. Mice were maintained on the same diet (eitehr CD or WD) for additional 5 weeks and tissues were collected for analysis.

Construction protocol: Trizol was used to extract total liver RNA Stranded RNA sequencing (RNAseq) libraries were constructed using Lexogen's QuantSeq 3' mRNA-Seq Library Prep Kit FWD for Illumina and the established protocols. The library construction was done using 500 ng of total RNA. Each of the libraries was indexed during library construction to multiplex for sequencing. Samples were normalized, and the libraries were pooled and run on Illumina's NovaSeq Platform
