We newly invented the EV sheet, which is a unique nanofiber paper to capture extracellular vesicles from around 10 uL of bio-fluids. In this study, we investigated miRNA profiles of tumor-surface ascites, serum, and urine using the EV sheet. Overall design: We attached the EV sheet to the moistened organs to collect a tiny amount of ascites. Moreover, the EV sheet was soaked in a tiny amount of ascites, serum, and urine. Then, total RNA was extracted from the EV sheet, and small RNA libraries were prepared. Finally, a single-end sequence was performed. Similarly, tissue miRNA profiles were obtained.

Construction protocol: The EV sheet was fabricated from cellulose nanofibers. The EV capture process from moistened organs using the EV sheet consists of the following steps: EV capture by absorption of ascites on organs, EV storage by drying, washing, and EV release. Then, total RNA was extracted using RNA purification kit (Norgen Biotek Corp, Thorold, Canada). To obtain tissue RNA, tissues were shredded using the gentleMACS Dissociator (Miltenyi Biotec, Bergisch Gladbach, Germany) and total RNAs was extracted by using the miRNeasy Plus Mini Kit (QIAGEN, Hilden, Germany). Small RNA libraries were prepared using the NEBNext Multiplex Small RNA Library Prep Set for Illumina (New England Biolabs, Ipswich, MA). Then, PCR products were separated by electrophoresis (120 V, 60 min), and DNA fragments corresponding to 140–160 bp (the length of small non-coding RNA plus the adaptors) were recovered.
