Study: Genome-wide maps of ESR2 chromatin state in Mantle cell lymphoma cells

In order to elucidate chromatin binding regions of ESR2 in Mantle cell lymphoma, we used ESR2 antibodies, opti-mized chromatin-immunoprecipitation (ChIP), and performed ChIP-Seq of ESR2 over-expressed Granta-519 cells and Granta519-mock cells.We present for the first time, the cistrome of nuclear receptor ESR2 in Mantle cell lymphoma cell line. Overall design: Examination of ESR2 chromatin binding in ESR2 over-expressed Granta-519 cells and Granta519-mock cells. Each ChIP was performed twice.

Construction protocol: Cells were crosslinked with 1% formaldehyde for 10 minutes at room temperature and nuclear chormatin fractions were separated and sonicated, followed by immunoprecipitated with ESR2 anitboides (PPZ0506, R&D systems). Magnetic beads were used to pull down the immunoprecipitated DNA and separated by elution buffer containing proteinase K. Then ChIP DNA was purified with QIAquick PCR Purification kit (Qiagen). The libraries of ChIP DNA were prepared by using the kit NEB Next Ultra II DNA Library Prep Kit for Illumina following user instrcutions
