Assay for transposase-accessible chromatin-sequencing (ATAC-seq) was performed in a time-series of cardiac differentiation of mESCs Overall design: Two biological replicates were collected every 24 hours of cardiac differentiation of A2lox.cre mESCs from the 2i stage through day 8, in addition to day 10 and day 15 (cardiomyocyte stage). Samples were then lysed, and transposition was carried out at 37C for 30 minutes with Illumina's Nextera DNA Library Prep kit (Illumina 15028212). Libraries were generated from transposed DNA and sequenced on an Illumina HiSeq4000 instrument in the University of Chicago's Genomic Facility.
