Design: RNA-seq and library preparation were performed at the University of Wisconsin Biotechnology Center DNA Sequencing Facility. Poly-A selection was employed to enrich high quality mRNAs from total RNA samples (1 micro gram input per sample) and library preparation was accomplished using the Illumina Truseq Stranded mRNA preparation kit (Illumina Inc.). After PCR enrichment of DNA fragments with adapter molecules, an Agilent DNA 1000 Chip (Agilent Technologies) assay was performed to verify the librarys profile and size.

Obesity, an immense epidemic affecting approximately half a billion adults, has doubled in prevalence in the last several decades. Epidemiological data support that obesity due to intake of a high-fat, western diet increases the risk of colon cancer; however, the mechanisms underlying this risk remain unclear. Here, utilizing next generation RNA sequencing, we aimed to determine the high-fat diet mediated gene expression profile in mouse colon and the AOM/DSS model of colon cancer.
