Design: To assess the efficiency of CRISPR/CAS9 indel formation using three gRNAs targeting 9-cis-EPOXYCAROTENOID DIOXYGENASE4 (LsNCED4), amplicons of LsNCED4 were generated from two lettuce (Lactuca sativa) cultivars: Cobham Green and Salinas. These amplicons were sequenced on Illumina HS3000 or HS4000 using a Nextera dual indexing strategy. Provided samples are already demultiplexed. Sample names are Cultivar_gRNA_plant#. T1 samples for gRNA 1 were amplified with primers NCED4_gRNA1_F/NCED4_gRNA1_R; T1 samples for gRNA 2 and gRNA 3 were amplified with primers NCED4_gRNA2+3_F/ NCED4_ gRNA2+3_R. All T2 samples were amplified with primers LDB_226/LDB_227.

This work describes a detailed, high resolution analysis of editing events in lettuce. This provides insights into mutational events resulting in stable transformants as a foundation for numerous further editing experiments in lettuce and other species. These editing events of the NCED4 gene are a phenocopy of a naturally occurring allele and result in a phenotype that is observable at the whole plant level with minimal pleotropic consequences. Therefore, editing of NCED4 could be used in a co-editing strategy to enrich for germline editing events.
