Construction protocol: Chicken bone marrow derived macrophage (BMDM) culture: RNA from chicken BMDM, day 7 embryos, or DF-1 cells, was extracted using TRIzol reagent, as described by the manufacturers (Invitrogen). RNA was quantified using a Nanodrop and RNA quality was assessed using Agilent nanochip Samples were prepared for mRNA sequencing using 1ug of total RNA starting material following the Illumina Truseq RNA Sequencing protocol. Resulting libraries were quality checked on an Agilent DNA 1000 bioanalyzer (Agilent Technologies, South Queensferry, UK) and then clustered onto a paired read flowcell using the Illumina v2 cluster generation kit at a 8pM concentration.
