In this study we performed single-cell sequencing of E13.5 mouse cerebella, revealing different newly generated neurons and their associated molecular features. Overall design: Design: Single-cell suspensions were prepared using Accumax digestion followed by manual trituration using a Pasteur pipette. Cells were processed with the 10X Genomics Chromium Single Cell Kit Version 1

Construction protocol: The cerebella were dissected from 7 E13.5 mouse embryos in ice-cold phosphate buffered saline. The dissected tissues were pooled and cut into pieces smaller than 1 mm in dimension and digested with Accumax in a 1.5-ml tube at 37˚C for 5-10 min. At the end of digestion, the tissue pieces were dissociated by gentle trituration with a wide-bore pipet tip. The cell suspension was filtered with a 100-µm cell strainer. After single cell suspension, the 10X Genomics Chromium Single Cell Kit v1 (PN-120233) was used to create libraries.
