Construction protocol: Dermal wound compartment was microdissected, and single cell suspension was created by incubating tissues in Dispase/Collagenase/Liberase. Dead cells were removed using Dead Cell Removal Kit, counted using Countess, and loaded into the Chromium platform (10X Genomics). Single cells were captured using the Chromium Platform (10X Genomics) and libraries were generated using Single Cell 3' v2 chemistry. Library metrics were as follow: 550.19 pg/µl with an avg. size ~454 bps. Libraries were sequenced on an Illumina HiSeq 4000 platform (Illumina). Cell counting, capture, library preparation, quality control, and sequencing was performed at the Genomics High Throughput Sequencing facility at the University of California, Irvine.
