Design: We used the NEB Ultra II kit (#E7645L) with the 96 NEBNext Multiplex Oligos for Illumina primers (#E6609L). We made libraries and then pooled up to 4 libraries each for a targeted sequence capture using MycroArray baits MyBaits COS Compositae/Asteraceae 1kv1 (#305296), with a later amplification with KAPA Ready Mix and primers P5 and P7 (the reamp primers described in Meyer & Kircher 2010 (doi:10.1101:pdb.prot5448) for Illumina libraries).

We used a Hyb-Seq method based on a probe set of 1061 nuclear conserved orthology loci with the followingaims: (1) to obtain a well resolved phylogeny of subtribe Saussureinae with a complete genus sampling; and (2) to test the validity of the genera described or accepted in recent taxonomic treatments based on phylogenetic evidence.
