Recent work by the Cook and Misteli laboratories addresses the issue of assembly and action of the transcription machinery. Live cell studies of RNA polymerases I (RNAP1) and II (RNAP2) indicate that transcription complexes are assembled in a stochastic fashion from freely diffusible subunits. This contrasts to models involving stable holo-complexes.
