Cytosolic 5-prime nucleotidases, such as NT5C1B, catalyze production of adenosine, which regulates diverse physiologic processes (Sala-Newby and Newby, 2001).
Sala-Newby and Newby (2001) cloned mouse Nt5c1b, which encodes a 573-amino acid protein that shares 66% identity with pigeon and human NT5C1A (610525). Mouse Nt5c1b contains 4 putative kinase-3a sequences and 3 nucleoside monophosphate-binding type-2 sequences. By database analysis and RACE, Sala-Newby and Newby (2001) obtained a partial human NT5C1B cDNA. The partial human protein shares 87% identity with mouse Nt5c1b and 70% identity with human NT5C1A. RT-PCR analysis of human tissues revealed ubiquitous expression of NT5C1B, with highest level in testis and lowest levels in brain and skeletal muscle. Mouse Nt5c1b also showed highest expression in testis.
Using overexpression in COS-7 cells, Sala-Newby and Newby (2001) showed that mouse Nt5c1b was activated by ADP and catalyzed adenosine formation during ATP breakdown.
By genomic sequence analysis, Sala-Newby and Newby (2001) mapped the NT5C1B gene to chromosome 2.
