Cloning and Expression

Davis-Smyth et al. (1996) identified FBP2 (KHSRP; 603445) and FBP3 as proteins related to the far upstream element (FUSE)-binding protein FBP1 (FUBP1; 603444), which binds to the FUSE of the human protooncogene MYC (190080). All 3 proteins had the same general architecture, with 3 distinct domains including a central nucleic acid-binding region containing KH motifs. The FBP3 protein contains 600 amino acids. Northern blot analysis revealed that FBP3 was expressed as a 3.4-kb mRNA in all cells and tissues tested.


Gene Function

Using expression studies in mammalian cells, Davis-Smyth et al. (1996) found that the C-terminal domains of the 3 FBPs are potent transcriptional activators. Each FBP bound specifically to 1 strand of the FUSE of MYC in vitro.
