[PRB3] The protein encoded by this gene is a proline-rich salivary protein. It is a major constituent of parotid saliva. This protein is proposed to act as a bacterial receptor. This gene and five other genes that also encode salivary proline-rich proteins (PRPs), as well as a gene encoding a lacrimal gland PRP, form a PRP gene cluster in the chromosomal 12p13 region. RefSeq annotates one representative transcript (NM included in AceView variant.b), but Homo sapiens cDNA sequences in GenBank, dbEST, Trace and SRA, filtered against clone rearrangements, coaligned on the genome and clustered in a minimal non-redundant way by the manually supervised AceView program, support at least 4 spliced variants.

Expression: According to AceView, this gene is expressed at low level, only 15.7% of the average gene in this release. The sequence of this gene is defined by 20 GenBank accessions from 18 cDNA clones, some from head neck (seen 5 times), salivary gland (3), human embryonic stem cells (once). We annotate structural defects or features in 2 cDNA clones.
Alternative mRNA variants and regulation: The gene contains 7 distinct introns (5 gt-ag, 2 others). Transcription produces 4 alternatively spliced mRNAs. The mRNAs appear to differ by presence or absence of a cassette exon, overlapping exons with different boundaries, splicing versus retention of one intron.
2 variants were isolated in vivo, despite the fact that they are predicted targets of nonsense mediated mRNA decay (NMD).
Function: There are 9 articles specifically referring to this gene in PubMed. Proteins are expected to have molecular function (Gram-negative bacterial cell surface binding) and to localize in various compartments (extracellular region, extracellular space). A putative protein interactor has been described (ALB).
Protein coding potential: The 4 spliced mRNAs putatively encode good proteins, altogether 4 different isoforms (2 complete, 2 partial); the 2 complete proteins appear to be secreted, apparently Homo sapiens specific.
