[MPP7] Membrane-associated guanylate kinases (MAGUKs) are important adaptor proteins involved in the assembly of protein complexes at sites of cell-cell contact. They are found in synapses, adherens junctions, and tight junctions. All MAGUKs contain at least 1 PDZ domain, an SH3 domain, and a GUK domain, and many contain 1 or 2 L27 domains, which are involved in multimerization of MAGUKs. MPP7 belongs to the p55 stardust subfamily of MAGUKs, which is named for a Drosophila gene required for establishment of cell polarity in the developing fly embryo (Bohl et al., 2007 [PubMed 17237226]).[supplied by OMIM].

RefSeq annotates one representative transcript (NM included in AceView variant.a), but Homo sapiens cDNA sequences in GenBank, dbEST, Trace and SRA, filtered against clone rearrangements, coaligned on the genome and clustered in a minimal non-redundant way by the manually supervised AceView program, support at least 12 spliced variants.

Expression: According to AceView, this gene is well expressed, 1.0 times the average gene in this release. The sequence of this gene is defined by 115 GenBank accessions from 106 cDNA clones, some from testis (seen 17 times), uterus (8), placenta cot 25-normalized (7), leiomyosarcoma (6), brain (5), pooled germ cell tumors (5), breast carcinoma (4) and 43 other tissues. We annotate structural defects or features in 2 cDNA clones.
Alternative mRNA variants and regulation: The gene contains 33 distinct gt-ag introns. Transcription produces 12 alternatively spliced mRNAs. There are 5 probable alternative promotors, 6 non overlapping alternative last exons and 8 validated alternative polyadenylation sites (see the diagram). The mRNAs appear to differ by truncation of the 5' end, truncation of the 3' end, presence or absence of 5 cassette exons, overlapping exons with different boundaries.
2 variants were isolated in vivo, despite the fact that they are predicted targets of nonsense mediated mRNA decay (NMD).
Efficacy of translation may be reduced by the presence of a shorter translated product (uORF) initiating at an AUG upstream of the main open reading frame (in variant aAug10, iAug10).
Function: There are 5 articles specifically referring to this gene in PubMed. Functionally, the gene has been tested for association to a disease (Alzheimer Disease). Proteins are expected to have molecular function (protein binding) and to localize in various compartments (adherens junction, cell junction, membrane, tight junction). A putative protein interactor has been described (AMOT).
Protein coding potential: 10 spliced mRNAs putatively encode good proteins, altogether 8 different isoforms (3 complete, 1 COOH complete, 4 partial), some containing domains Guanylate kinase, L27 domain, PDZ domain (Also known as DHR or GLGF), SH3 domain, Variant SH3 domain [Pfam]. The remaining 2 mRNA variants (2 spliced; 2 partial) appear not to encode good proteins.
Isoform MPP7.dAug10 is annotated using as Met a Kozak-compatible g..GTGg start, thereby gaining 161 amino acids N-terminal to the first AUG. Finally proteins from this gene may be modulated by acetylation; phosphorylation, as detailed at PhosphoSite.
