RefSeq annotates one representative transcript (NM included in AceView variant.c), but Homo sapiens cDNA sequences in GenBank, dbEST, Trace and SRA, filtered against clone rearrangements, coaligned on the genome and clustered in a minimal non-redundant way by the manually supervised AceView program, support at least 29 spliced variants.

Note that this locus is complex: it appears to produce several proteins with no sequence overlap.
Expression: According to AceView, this gene is expressed at high level, 3.4 times the average gene in this release. The sequence of this gene is defined by 645 GenBank accessions, some from brain (seen 52 times), hippocampus (17), lung (16), thymus (14), testis (12), uterus (12), amygdala (11) and 129 other tissues.
Alternative mRNA variants and regulation: The gene contains 30 distinct introns (27 gt-ag, 3 gc-ag). Transcription produces 34 different mRNAs, 29 alternatively spliced variants and 5 unspliced forms. There are 4 probable alternative promotors, 5 non overlapping alternative last exons and 6 validated alternative polyadenylation sites (see the diagram). The mRNAs appear to differ by truncation of the 5' end, truncation of the 3' end, presence or absence of 15 cassette exons, overlapping exons with different boundaries, splicing versus retention of 2 introns. 165 bp of this gene are antisense to spliced gene C5orf4.aAug10, raising the possibility of regulated alternate expression.
Note that mRNA .vdAug10 was found in vivo, although it is a predicted target of nonsense mediated mRNA decay (NMD).
Efficacy of translation may be reduced by the presence of a shorter translated product (uORF) initiating at an AUG upstream of the main open reading frame (in variant cAug10, eAug10).
Function: There are 11 articles specifically referring to this gene in PubMed. Functionally, the gene has been tested for association to a disease (Myelodysplastic Syndromes) and proposed to participate in pathway (RNA degradation). Proteins are expected to have molecular function (nucleic acid binding) and to localize in various compartments (cytoplasm, extracellular space, mitochondrion). Putative protein interactors have been described (BTG1, BTG2, CNOT1, CNOT2, CNOT3, CNOT4, CNOT6, CNOT10ANDSUGT1P2, MCM7).
Protein coding potential: 26 spliced mRNAs putatively encode good proteins, altogether 20 different isoforms (9 complete, 1 COOH complete, 10 partial), some containing CAF1 family ribonuclease domain [Pfam]; 2 of the 9 complete proteins appear to be secreted. The remaining 8 mRNA variants (3 spliced, 5 unspliced; 4 partial) appear not to encode good proteins.
2 isoforms are annotated using as Met a Kozak-compatible non-AUG start, thereby gaining a minimum of 99 amino acids N-terminal to the first AUG. Finally proteins from this gene may be modulated by phosphorylation; ubiquitination, as detailed at PhosphoSite.
