Treatment protocol AMG was added to the medium, when needed
Growth protocol pAstros from different genotypes were transduced with the indicated retroviruses; 48 hours later, the medium was changed with differentiation medium.

Extraction protocol Cells were collected and FACS sorted at 5 days post infection, each sample into a separate tube. Then, CMO solution was added to the cells, according to manifacturer's protocol (10XGenomics).
Equal number of cells from each condition was added to have a correct cell concentration. Then, ~20000 cells were loaded per lane, and 2 lanes were loaded.
Libraries were prepared according to manifacturer's protocol (10XGenomics).
