Growth protocol 10% FBS, 1% penicillin/streptomycin, high glucose DMEM
Extracted molecule polyA RNA
Extraction protocol About 10e6 4T1 cells were isolated from different group and the mRNA library constructed using QIAseq FX Single Cell RNA Library Kit (QIAGEN) according to the manufacturer’s instruction
RNA libraries were prepared for sequencing using standard Illumina protocols

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina HiSeq 2000

Data processing Illumina Casava1.7 software used for basecalling.
Sequenced reads were trimmed for adaptor sequence, and masked for low-complexity or low-quality sequence, then mapped to mm10 whole genome using bowtie v0.12.9.
Fragments Per Kilobase of exon model per Million mapped fragments (FPKM) was performed with Cufflinks v2.1.1, aligned reads were counted with HTSeq.
