Extraction protocol |
RNA was extracted using RNeasy Micro Kit (Qiagen) according to protocol (neuronal Bchs overexpression dataset) or exchanging the first step of homogenization in RLT buffer with the following Trizol protocol (glial Bchs overexpression dataset). Heads were homogenized in 500 µl Trizol, centrifuged for 5 min at 10000 rpm at 4°C, supernatant was tranferred to a new tube, 200 µl Chloroform (Carl Roth, No. 6340.1) was added and the tube was shaked. An incubation step at room temperature for 3 min was followed by centrifugation for 5 min at 10,000 rpm at 4°C. The upper aqueous phase was transferred in a new tube and it was continued with the RNeasy Micro Kit protocol. RNA-seq libraries were prepared using TruSeq RNA Library Prep Kit v2 (Illumina, San Diego, CA).
