Extraction protocol Total RNA of wild type or mcm5 mutated zebrafish at 3.5dpf was extracted, and mRNA was enriched by olig(T) beads.
Add Fragmentation Buffer to randomly break mRNA into short fragments. Reverse transcription synthesis of cDNA using short fragments as templates, and then perfom end repair and A-tail addition for all cDNA. Following add UMI tags and sequencing adapters onto the cDNAs. Finally, PCR amplification was performed to obtain the library.. All the DNA libraries were sequenced by HiSeq X Ten. And the clean data were generated from the raw data by removing the adapters.
