Preparation: collect the lymph nodes from the control (n=3), RHOAG17V (n=1), and RHOAG17V; DNMT3AR882H AITL (n=1) group. The samples were washed with 2% (v/v) FBS-PBS. Filtration of ground lymph node tissue through a 100um filter to form a single-cell suspension. After quantifying the cell number, adjusting the concentration to 800-1200 cells/μl, and ensuring the cell activity > 85%, single-cell suspensions from these specimens were subjected, typically within an hour after the acquisition, to single-cell RNA sequencing (scRNA-seq) using the 10X genomics platform to construct the single-cell library of samples.
