Extraction protocol RNA was harvested using RNeasy Micro Kit(Qiagen). 12ng of total RNA was used for the construction of sequencing libraries.
RNA libraries for RNA-seq were prepared using SMART-Seq Stranded Kit following manufacturer's protocols.

Data processing The quality of the bulk RNA-seq reads was assessed using FastQC (v0.11.5).
The reads were aligned to the mouse reference genome (mm10) with Bowtie2 (v2.3.3.1) using RSEM (v1.3.0) with the default parameters. The function rsem-calculate-expression was used to align the reads and quantify the gene and isoform abundance.
