Growth protocol |
Primary NSCs were isolated from the brain SVZ of the young (2 months) and aged (15-18 months) mice, respectively (Nat Protoc. 2012 Nov; 7(11):2005-2012). Briefly, microdissection of the brain SVZ was performed on ice. The tissue pieces of SVZ from 5 mice were pooled and dissociated into single cells using Neural Tissue Dissociation kit. The isolated NSCs were maintained in the initial proliferation medium.

Total RNA extraction of the primary NSCs was performed using TRIzol Reagent following manufacturer’s instructions (Thermo Fisher). 1 ug of total RNA was used for rRNA depletion (rRNA depletion kit, MGI) and RNAseq library generation using MGIEasy RNA Library Prep Set (Item No.:1000006383)

Data processing |
RNAseq reads were aligned to mm10 genome using TopHat with default parameters. Aligned reads were converted to sam format. Cuffdiff software was used to determine the transcript FPKM (Fragments Per Kilobase of transcript per Million mapped reads). Assembly: mm10 Supplementary files format and content: tab-delimited text files include FPKM values for each sample.
