Treatment protocol |
TFK-1 cells were transduced with specific lentiviruses at a multiplicity of infection (MOI) of 20 in the presence of HistransG (Genechem) for 24 h. The cells were cultured in complete medium for two days and treated with 1 μg/ml of puromycin for the establishment of stable cell lines.

TFK-1 was maintained in RPMI-1640 with 10% FBS.

Extraction protocol |
Total RNA was extracted from individual cell samples using RNA Extraction Reagent (Vazyme) according to the instructions. The samples were resuspended with nuclease-free water.RNA quantity and purity was assessed using NanoDrop ND-1000.

Target preparation was performed using an Eberwine-based amplification method with Amino Allyl MessageAmp II aRNA Amplification Kit (Ambion, AM1753) to generate amino-allyl antisense RNA(aa-aRNA). Cy5-labeled aRNA coupled with NHS-CyDye was prepared and purified prior to hybridization. Purified coupled aRNA was quantified using NanoDrop ND-1000.

Hybridization protocol |
10 ug of Cy5-labeled aRNA was hybridized to the Phalanx Human OneArray 6.1 by Phalanx Hybridization System according to the Phalanx Hybridization Protocol.

Scan protocol |
After hybridization, the arrays were washed, scanned and the gene expression results were extracted by DNA Microarray Scanner G2565B (Agilent), according to the manufacturer’s instructions.
