Status Public on Nov 18, 2010
Title 2 Wild-type control macrophage+RSG
Sample type RNA

Source name Wild-type bone marrow-derived macrophage treated with RSG
Organism Mus musculus
Characteristics mouse: 2
strain: C57Bl/6
genotype/variation: Wild-type
cell type: bone marrow-derived macrophage
activation: Control
treatment: Rosiglitazone
time: 10 days
Treatment protocol Cell were treated after plating into 6-well cell culture plates with the indicated cytokines (20 ng/ml M-CSF. 20 ng/ml IL-4 and vehicle (DMSO:ethanol) or 1 uM Rosiglitazone.
Growth protocol Bone marrow cells were isolated from the femur, washed twice with PBS and cultured in RPMI+10% FCS supplemented with glutamine, penicillin and streptomycin and 20 ng/ml M-CSF. Fresh media and cytokines were added every other day for 10 days.
Extracted molecule total RNA
Extraction protocol Qiagen extraction of total RNA was performed according to the manufacturer's instructions.
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 5 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol Following fragmentation, 10 ug of cRNA were hybridized on Mouse Genome 430 2.0 Affymetrix microarrays according to Affymetric standardized protocol.
Scan protocol Microarrays were scanned canned according to Affymetric standardized protocol.
Description Gene expression data of RSG-treated bone marrow-derived macrophages derived from C57Bl/6 wild-type mouse
6_(Mouse430_2).CEL
Data processing The data were generated with GCOS, and cel files were analyzed in GeneSpring 7.3 using GC-RMA.
