Treatments represent the FT1 and FT2 under both constitutive overexpression and heat shock promotion, as well as the respective controls.

Growth protocol |
For 35S samples, leaf tissues were collected at approximately 9 AM. For HSP promotion treatments, leaf samples were harvested at day 21 before the end of heat shock treament. All plants were of the approximate same developmental stage and were about 1 to 2 years old.

RNA was extracted with a hot-borate method (Wan and Wilkins; 1994), followed by a Dnase I digestion and clean-up using the Rneasy Mini Kit (Quigen, Valencia, CA).

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 3 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45C on a GeneChip Poplar Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using a GeneChip Scanner 3000 (Affymetrix).
