Extraction protocol Hematopoietic organs were dissected and dissociated into single cells with collagenase type I (Sigma, C0130, 0.12% v/v)
AGMs from NG2 +/+; Runx1 fl/fl and NG2-Cre; Runx1 fl/fl embryos were dissected, dissociated, washed, counted and assessed for viability using a Bio-Rad TC20. Cells were then loaded at a concentration for the recovery of 7,000 cells and processed through the 10x Chromium Controller. Single cell libraries were obtained according to manufacturer protocol. RNA concentration was obtained using Quibit RNA HS (Thermo-Fisher) and the quality of the libraries was verified using the LabChip GX24 (Perkin Elmer).
