Bone marrow derive dendritic cells were routinely generated by re-plating d9 bone marrow cell differentiation cultures (typically 2–3 × 106 cells into 100mm diameter dish) on uncoated plastic in complete RPMI1640 (Gibco) supplemented with 20 ng/ml of GM-CSF (PeproTech).

BMDCs were harvest, live CD11c hi BMDCs were sorted into lysis buffer, flash frozen on dry ice. Illumina TruSeq RNA Sample Prep Kit (Cat#FC-122-1001) was used with 1 ug of total RNA for the construction of sequencing libraries. RNA libraries were prepared for sequencing using standard Illumina protocols
