Extraction protocol Total RNA from placentas was isolated using RNeasy Mini Kits according to the manufacturer’s instructions. RNA quantity was determined using an Agilent Bioanalyzer.Total RNA from placentas was purified using mirVanaTM miRNA Isolation Kits and quantified.
Label biotin
Label protocol Starting from Total RNA, poly A tail was attached to the 3 'end of the small RNA under the guidance of poly A polymerase. Under the guidance of ligase, biotin labeled patented 3DNA Dendrimer was attached to the poly A tail of RNA.
 
Hybridization protocol Before using 20X Eukaryotic Hybridization Controls for the first time, confirm whether denatured at 65℃ for 5min has been performed. The hybridization solution was incubated in a constant temperature metal bath at 99°C for 5 min, and then 45°C for 5 min. The hybrid solution was removed from the thermostatic metal bath and centrifuged for 5 min at 13200rpm in a miniature centrifuge to remove insoluble substances in the hybrid mixture. Hybridized was preformed in a Agilent Hybridization Oven for 16hr at a rotation speed of 60 rpm at 48℃.
