Cells were left untreated (0h), or treated with 250nM 4OHT (2h, 8h), or treated with 4OHT for 8hrs followed by a subsequent 2h Mek inhibitor treatment (8_2h).

mESCs were grown in serum containing medium, supplemented with 1x LIF (made in house) and 2uM Chir99021. 24hrs prior to treatment, media was changed to serum containing media supplemented with 1x LIF and 250nM PD173074.

Cells we fixed in 1% formaldehyde at RT for 10 minutes, nuclei extracted and chromatin sonicated using a bioruptor (10 cycles, 30s on 30s off). Cleared chromatin was incubated with anti-RBP1 antibody (Active Motif, 39097). Input DNA serves as negative control for antibody enrichment. Libraries were prepared using the NEB Next Ultra 2 kit.
