Tfh cells (SLAM-loCD44+CD62L-CD4+) were directly sorted into TRIzol reagent by a FACSAriaII sorter, and total RNA was extracted using standard Qiagen RNease columns. RNA libraries were prepared for sequencing using standard Illumina protocols。

The quality of RNA-Seq libraries was assessed by FastQC (v0.11.5). RNA-Seq libraries were mapped to mouse genome mm10 (version M17) using TopHat (v2.1.1). Expression levels of all genes were estimated by HTseq (v0.6.1), and Reads Per Kilobase per Million mapped reads (RPKM) counts were calculated.
