All mice in this study were kept in C57BL/6J backgroud. Pro-/pre B cells were sorted from Rosa26BirA/BirA transgenic mice transduced with virus over-expressing bio-tagged Hoxb5 gene (day 3).

Sonication and Chromatin Immunprecipitation (Chip) by dynabeads M-280 Streptavidin antibody (Thermo Fisher, catalog is 11205D,2 mL, lot:00381796), and genomic DNA was extracted by Phenol chloroform method. Chipseq libraries were constructed using VAHTSTM Universal DNA library Prep Kit for illumina (ND604) and libraries sequence procedure performed on HiSeq X Ten system.

Data processing |
Chipseq fastq files were aligned to mm10 genome by bowtie2. The generated bam files werer used as the input files for Homer packages. Finding Hoxb5 binding peaks and annotation information procedure was according to the Homer user manual with default parameters for transcription factor.
