Treatment protocol |
Fenofibrate was dissolved in 0.5% hydroxypropyl methylcellulose (Shinestu Kagaku Kogyo, Japan) and administered orally to the mice at a dose of 300 mg/kg once per day for 4 days with a disposable plastic sonde (Fuchigami Kikai Co., Kyoto, Japan). Three mice received fenofibrate and three mice received the control treatment with vehicle only.

The animal study was carried out in PXB mice (genotype: cDNA-uPA+/wt/SCID, uPA+/wt; B6;129SvEv-Plau, SCID; C.B-17/Icr-scid/scid Jcl) at PhoenixBio Co. Ltd. Briefly, cryopreserved human hepatocytes from a 2-year-old Hispanic girl were purchased from BD Biosciences (Woburn, MA, USA). After thawing, the hepatocytes were transplanted into hemizygous 2- to 4-week-old cDNA-uPA/SCID mice via the spleen under anesthesia. . A full description of the generation of these mice can be found elsewhere [PMID: 26536627]. Six male mice between 12-18 weeks of age were used for the study.

Extraction protocol |
Total RNA was prepared from liver using TRIzol reagent, whereafter purified total RNA was isolated using Qiagen RNEasy columns. RNA integrity was checked on chip analysis (Agilent 2100 bioanalyzer, Agilent Technologies, Amsterdam, the Netherlands) according to the manufacturer's instructions. RNA was judged as suitable for array hybridization only if samples exhibited intact bands corresponding to the 18S and 28S ribosomal RNA subunits, and displayed no chromosomal peaks or RNA degradation products (RNA Integrity Number > 8.0)

Purified total RNA (100ng per sample) was labeled with the Whole-Transcript Sense Target Assay (Affymetrix, Santa Clara, CA, USA; P/N 900652).

Hybridization and washing of the Affymetrix GeneChip Human Gene 1.1 ST peg arrays were performed on a GeneTitan Instrument (Affymetrix, Santa Clara, CA) according to the manufacturer's recommendations.
