Characteristics |
cell type: ES-derived neural stem cells treatment: Testosterone 100nM 4 hours antibody: AR (N-20 Santa Cruz sc-816)
|
Treatment protocol |
Cells were placed in depleted medium (phenol-red free medium without N2 and B27) during 48h hours before androgen treatments and thus treated with DMSO, testosterone, DHT 10nM or 100nM during 4h hours.
|
Growth protocol |
ES cell–derived hNSC cells were routinely maintained on poly-ornithine and laminin-coated dishes in Dulbecco's modified Eagle's medium/F12 Glutamax and Neurobasal medium (1:1) supplemented with N2, B27 and 50µM 2-mercaptoethanol containing brain-derived growth factor (20ng/mL), fibroblast growth factor-2 (10ng/mL) and epidermal growth factor (10ng/mL).

Extracted molecule |
genomic DNA |
Extraction protocol |
Cellular lysates were isolated by incubating cells pellets in SDS lysis buffer, sonicated and AR-DNA complexes were isolated with antibody. Libraries were prepared using NEXTflex ChIP-Seq Kit (#5143-02, Bioo Scientific) following the manufacturer's protocol (V12.10)
