Treatment protocol |
Cells were transfected with each TET1 knockdown vector or a pcDNA6.2-GW/EmGFP-miR-neg control plasmid (Thermo Fisher Scientific), after which they were selected with 0.6 mg/ml G418. Established clones were treated with our without with 1 μM 5-aza-2’-deoxycytidine (5-aza-dC, Sigma-Aldrich) for 72 h, replacing the drug and medium every 24 h.

Growth protocol |
HCT116 cells were cultured in McCoy's 5A medium supplemented with 10% heat-inactivated fetal bovine serum. The cultures were incubated at 37 ºC in a humidified incubator with 5% CO2.
