Treatment protocol Zygotes were injected with either tRF-GlyGCC oligo with modified bases (tRF-GlyGCC), tRF-Gly-GCC antisense oligo (tRF-GlyGCC Antisense), H3.3-GFP mRNA(GFP), tRF-GluCTC (tRF-GluCTC) or sperm small RNA from control or low protein animals.
Growth protocol Zygotes, generated by IVF using sperm from mice fed on control diet and oocytes from females on control diet, were microinjected 2 hours after IVF and then allowed to develop until late 2C stage in KSOM media. 32 hours post IVF the microinjected embryos were collected and frozen at -80C.
Extracted molecule total RNA
Extraction protocol 2C embryos positive for GFP signal were individually collected at 2C or 4C stage in 5ul of buffer TCL with 1% beta mercaptoethanol and frozen at -80C.
