| Title |
cultured PBMC from MS patient stimulated with PHA |
| Sample type |
RNA |
| |
|
| Source name |
cultured MS PBMC with PHA
|
| Organism |
Homo sapiens |
| Characteristics |
tissue: PBMC disease state: MS patient gender: female age: 40
|
| Treatment protocol |
To induce PBMC activation, Phytohemagglutinin (PHA) (ThermoFisher) (0.5%) was used.
|
| Growth protocol |
PBMCs were isolated from whole blood and cryopreserved in liquid nitrogen until used. After thawing, PBMCs were cultured in a P48 plate (200.000cells/well) for 72h.
|
| Extracted molecule |
total RNA |
| Extraction protocol |
miRNeasy mini kit (Qiagen)
|
| Label |
biotin
|
| Label protocol |
Total RNA was labeled using FlashTag Biotin HSR RNA labeling kit (Genisphere)
|
| |
|
| Hybridization protocol |
GeneChip Hybridization, Wash and Stain kit was used for hybridization in a GeneChip Hybridization oven 640 and in a GeneChip Fluidics Station 450 using the protocol FS450-0002, as explained in the protocol provided by Affymetrix.
|
| Scan protocol |
Arrays were scanned in a GeneChip 7G scanner and analysed in Affymetrix GeneChip Command Console Scan Control v4.0.0.1567.
|
| Description |
03-029EM_PHA
|
| Data processing |
Quality control, normalization and summarization of the data were performed in Expression Console software build 1.4.0.38 (Affymetrix). Only human probes.
