Treatment protocol |
iPSCs were generated via Sendai viral reprogramming of CD1 and Nos3-/- tail-tip fibroblasts. Cells were cultured on inactive MEF feeders in the pluripotent state and spontaneously differentiated via aggrewell-mediated EB formation to D11.

Growth protocol |
The study samples consisted of iPSCs (D0) reprogrammed from tail-tip fibroblasts of adult male wild type CD1 and Nos3-/- mice and differentiated embryoid bodies (D11).

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 100 ng total RNA.

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on Affymetrix GeneChip Mouse Genome 430 2.0 Arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the GeneChip Scanner 3000 7G.
