Treatment protocol Nicotine self-administration (intravenous infusion, 0.01, 0.03, 0.1, 0.25, 0.4 mg/kg/infusion), using an operant self-administration chamber, was conducted in the sound attenuating cubicle (Med Associates, Inc.) in the dark room.
Growth protocol C57BL/6 mice were individually housed for 12 hours with a reversed light-dark cycle in a laboratory breeding room.
Extracted molecule total RNA
Extraction protocol Total RNA was isolated using TriZol as described by the manufacturer (Ambion). RNA quality was assessed by Agilent 2100 bioanalyser using the RNA 6000 Nano Chip (Agilent Technologies, Amstelveen, The Netherlands), and quantity was determined by ND-1000 Spectrophotometer (NanoDrop Technologies, Inc., DE, USA).
Label biotin
Label protocol Per RNA sample, 1 ug were used as input into the procedure as recommended by protocol (Genisphere FlashTagTM Biotin RNA Labeling for Affymetrix GeneChip miRNA arrays). 1 ug of total RNA was used as input into the labeling reaction.
 
Hybridization protocol Labeled miRNA was hybridized to the array for 16 hours at 48 “C and 60 rpm as described in the protocol.
Scan protocol After hybridization, the chips were stained and washed in a Genechip Fluidics Station 450(Affymetrix) and scanned by using a Genechip Array scanner 3000 7G (Affymetrix).
Description miRNA
Data processing The scan image data was extracted through Affymetrix Commnad Console software1.1. The raw .cel file generated through above procedure meant expression intensity data and was used for the next step. Expression data were generated by Affymetrix Expression Console software version1.1. For the normalization, RMA (Robust Multi-Average) algorithm implemented in Affymetrix Expression Console software was used. Affymetrix miRNA-4 Array contains multispecies. The data of Mus musculus was analysed in this study.
