Treatment protocol Rats were exposed to isoflurane in oxygen at a concentration of 2% (1.4 MAC) or pure oxygen for 15 minutes and allowed to recover for 6 hours. Brain was removed and cortex dissected and placed into RNA later (Invitrogen, Paisly, UK) for RNA preservation.
Growth protocol 20 week old Sprague Dawley rats between 250-300g
Extracted molecule total RNA
Extraction protocol Total RNA was extracted using TRIZOL (Invitrogen, Paisly, UK). Total RNA was further purified using Quiagen column based methodology (Quiagen Ltd, Manchester, UK)
Label biotin
Label protocol Biotinylated cRNA was prepared according to Affymetrix standard protocol using 10ug total RNA
 
Hybridization protocol Following fragmentation, 10ug of cRNA were hybridized for 16 hours @ 45oC to rat 230 2.0 genechips
Scan protocol Following hybridization and staining, gene chips were scanned using at 3mm resolution using a gene array scanner

Data processing The data were analysed using Affymetrix microarray software suite 5.0 (MAS 5.0) using default analysis setting and global scaling as normalization method and Dchip ver 2010 using a baseline array for normalization and model based expression
