Growth protocol |
Cells were seeded in 48- and 6-well plates (Nunc) at 25,000 and 100,000 cells per well, respectively. Next day, the medium was replaced with adipocyte induction medium containing 1 µM Dexamethasone (Decadron), 0.5 mM iso-butyl-methyl-xanthine and 50 µM Indomethacine (both from Sigma-Aldrich). The differentiation medium was replaced twice a week for 21 days.

RNA was isolated using the standard TRIzol procedure (Invitrogen), and further purified with an RNeasy mini column (Qiagen GmbH, Hilden, Germany), according to the manufacturers’ instructions.
