RPMI medium + 10% FCS and supplements (Hepes, 2-Me, glutamine, NEAA, Na-Pyruvate, Pen/Strep); isolated primary cells stimulated as indicated at a density of 0.5million/mL.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 250 ng total RNA.

Hybridization protocol |
Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45C. on GeneChip Mouse Genome 430 2.0 Array. GeneChips were washed and stained, using an anti-biotin amplification step, in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the Affymetrix GeneChip® Scanner 3000 according to manufacturer's specifications.

Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings. The resulting data was then normalized and scaled using D-chip (PM-only) model.
