Overall design |
2 male Foxp3-DTR-GFP were used for the study. Single cells suspension from BM and spleen were prepared, then enriched for CD4 by magneic beads and sorted by gating on CD4+TCRb+. GFP was used to differentiate between Tregs and CD4conv. CD4conv or Tregs cells from the same tissue were pooled from the two mice and hashtagged (total of 4 groups: Tregs BM/Tregs spleen/CD4 conv BM/CD4 conv spleen). The mice were not treated by DT.
