Summary |
In order to understand why blocking gephyrin phosphorylation via mutating serines 268 and 270 to alanine disrupts PV interneuron development in the hippocampus, we recorded spontaneous and elicited activity in developing MGE derived interneurons within the dorsal hippocampal CA1 pyramidal cell layer, then aspirated the contents of the cells to detect differences in the transcriptomic state between sexes and genotypes.

Overall design |
We collected acute slices from post-natal day 4 male and female GPHNWT and GPHNS268/270A mouse pups which were crossed to an Nkx2.1 Cre: Ai14 TdT background, allowing us to record electrophysiologically from TdTomato postive MGE-derived interneurons within the CA1 pyramidal cell layer, representing an enrichment of putative parvalbumin (PV) type basket cells.
